چکیده

DNA methylation plays an important role in animal development and gene regulation. In mammals, several genes encoding DNA cytosine methyltransferases have been identified. DNA cytosine-5-methyltransferase 1 has a role in the establishment and regulation of tissue-specific patterns of methylated cytosine residues. DNMT1 is constitutively expressed and is required for the maintenance of global methylation after DNA replication. Aberrant methylation patterns are associated with certain human tumors and developmental abnormalities. The human DNMT1 gene was localized on 19p13.3→p13.2 (elDeiry et al., 1991; Yen et al., 1992). In this study using somatic cell hybrid cells and a radiation hybrid panel we have mapped the gene DNMT1 to porcine chromosome 2, thus confirming the expected localization of the gene in the porcine genome.

Assignment of the porcine FUS1 gene to SSC13q21q22 by somatic cell and radiation hybrid panel mapping.

FUS1 is a candidate tumor suppressor gene organized in a 370 kb region which contains a set of 19 genes. The human FUS1 genomic sequence spans 3.3 kb containing 3 exons (NM_007275; Lerman and Minna, 2000). Overexpression of the FUS1 gene leads to G1 arrest and growth inhibition of lung cancer cells (Kondo et al., 2001). The human and the murine FUS1 genes were assigned to chromosome HSA3p21.3 a...

Mapping of porcine ESTs obtained from the anterior pituitary.

We report the physical mapping of porcine expressed sequence tags (ESTs) from anterior pituitary clones isolated by differential display PCR in a study using lines selected for reproduction. These ESTs were mapped using a somatic cell hybrid panel (SCHP) and a radiation hybrid panel (IMpRH) as follows (SCHP position, nearest marker on the RH map): SPARCL1 (8q23-q27, SSP1); ATF4 (5p11-p15, AC02)...

A candidate mammalian DNA methyltransferase related to pmt1p of fission yeast.

Trace levels of 5-methylcytosine persist in the DNA of mouse embryonic stem cells that are homozygous for null mutations in Dnmt1 , the gene for the one previously recognized metazoan DNA methyltransferase. This residual 5-methylcytosine may be the product of a candidate second DNA methyltransferase, Dnmt2, that has now been identified in human and mouse. Dnmt2 contains all the sequence motifs ...

Xenoduplex analysis--a method for comparative gene mapping using hybrid panels.

Somatic cell hybrid (SCH) panels and radiation hybrid (RH) panels are powerful resources for comparative gene mapping because gene assignments are made without the detection of genetic polymorphism as needed for linkage mapping. A frequently encountered problem, however, is that the gene specific primers may amplify homologous PCR products of equal length from the donor and recipient species of...

Effects of 5-aza-2ˈ-deoxycytidine and Valproic Acid on Epigenetic-modifying DNMT1 Gene Expression, Apoptosis Induction and Cell Viability in Hepatocellular Carcinoma WCH-17 cell line

Background: DNA molecule of the eukaryotic cells is found in the form of a nucleoprotein complex named chromatin. Two epigenetic modifications are critical for transcriptional control of genes, including acetylation and DNA methylation. Hypermethylation of tumor suppressor genes is catalyzed by various DNA methyltransferase enzymes (DNMTs), including DNMT1, DNMT2, and DNMT3. The most well chara...
